You would use it as we use "what's the matter?" or "what's wrong?" in English, and in informal conversations (the formal way is どうしました[か] ).
For example, imagine a mother that giving a chocolate candy to his son けんた. The kid loves chocolate, but for whatever reason, this time he surprisingly refuses it. The surprised mother might say "どうしたの、けんた?"
