The most obvious thing is that 約 (which is yaku, rather than yoku) comes before the thing you're estimating, where as kurai or gurai comes after it.
約一週間 about 1 week
一週間ぐらい about 1 week
Beyond that, くらい・ぐらい is more conversational than 約.
くらい also has other grammatical functions that 約 can't have, like 目に見えないくらい小さい (so small that it can't be seen with one's eyes).
